    Ideal time for taking Plaquenil and side effects? Lupus. I actually take all 400 mg in the morning after eating breakfast because after my stomach adjusted, I have tolerated the Plaquenil very well. My rheumy doesn't think there's a problem with taking it all at once as long as I'm tolerating it since there is such a long half-life, it shouldn't affect the drug itself.
